Valencia is about 1 h 54 to 2 h from Madrid by direct train, with 20 to 26 direct trains a day according to ticket aggregators, from about €25 (average same-day price around €52). Trains are run by three operators (Renfe's AVE, Ouigo, Iryo) and leave from Atocha or Chamartín depending on the company: check the station on your ticket. It is doable in a day, but a night there is more comfortable.
Highlights

The City of Arts and Sciences
Calatrava's futuristic complex and its pools: Valencia's signature photo, best in late afternoon when the light changes.
The historic centre
The cathedral, the Lonja de la Seda and the Mercado Central can be visited on foot in the same morning.
A paella near the Albufera
Paella was born around this lake: book a restaurant in that area rather than the centre for the traditional version.
